Caplyta (lumateperone) was reviewed by the FDA for the treatment of depressive episodes associated with bipolar I or II disorder (bipolar depression) in adults, both as monotherapy and as adjunctive therapy. Its review highlighted a statistically significant improvement in MADRS Total Score, with an LS mean difference versus placebo of -4.6. Vraylar (cariprazine) was reviewed for the treatment of depressive episodes associated with bipolar I disorder (bipolar depression) in adults, and also as adjunctive therapy to antidepressants for major depressive disorder (MDD). For bipolar depression, the review noted a MADRS placebo-subtracted change ranging from -1 to -2.5.
