What the regulatory reviews say

Enhertu (trastuzumab deruxtecan) is a HER2-directed antibody-drug conjugate reviewed for adult patients with unresectable or metastatic HER2-positive (IHC 3+) solid tumors who have received prior systemic treatment and have no satisfactory alternative treatment options, and for adult patients with unresectable or metastatic non-small cell lung cancer (NSCLC) whose tumors have activating HER2 (ERBB2) mutations and who have received a prior systemic therapy. The FDA review for Enhertu reported an Objective Response Rate (ORR) of 60.3% and a median Duration of Response (DoR) of 14.8 months.

Kadcyla (trastuzumab emtansine) is also a HER2-directed antibody-drug conjugate, reviewed for the treatment of adult patients with HER2-positive, unresectable locally advanced or metastatic breast cancer who previously received trastuzumab and a taxane. The EMA review for Kadcyla reported an IRC-assessed Objective Response Rate (ORR) of 32.7% and a median duration of objective response of 9.7 months.
