What the regulatory reviews cover
This library includes the PARP inhibitors Lynparza, AKEEGA, and Zejula, which have undergone regulatory review for various indications. Lynparza was reviewed for its use in combination with abiraterone and prednisone or prednisolone for the treatment of adult patients with mCRPC, and as monotherapy in patients with deleterious or suspected deleterious germline BRCA-mutated advanced ovarian cancer. AKEEGA was reviewed for metastatic hormone-sensitive prostate cancer (mHSPC) and BRCA 1/2 mutations, and with prednisone or prednisolone for metastatic castration-resistant prostate cancer (mCRPC) with BRCA1/2 mutations. Zejula was reviewed as monotherapy for the maintenance treatment of adult patients with platinum-sensitive relapsed high-grade serous epithelial ovarian, fallopian tube, or primary peritoneal cancer.
Different drugs were studied in different trials, so figures are not head-to-head comparisons. For Lynparza, the Hazard Ratio for rPFS (ITT population) was 0.66. AKEEGA demonstrated a Median rPFS (months) for niraparib + AAP arm (All HRR population) of 51.6 months, compared to 33 months for the AAP arm (All HRR population). Zejula showed an IC50 for PARP1 inhibition of 1.1 nM.
