Leqembi and Kisunla are anti-amyloid antibodies reviewed for the treatment of early Alzheimer's disease. Leqembi is indicated for adult patients with a clinical diagnosis of mild cognitive impairment and mild dementia due to Alzheimer’s disease (Early Alzheimer’s disease) who are apolipoprotein E ε4 (ApoE ε4) non-carriers or hetero. The regulators' review of Leqembi noted a change from baseline in brain amyloid plaque (SUVR) of -0.310. Kisunla is indicated for the treatment of adult patients with a clinical diagnosis of mild cognitive impairment and mild AD dementia due to Alzheimer’s disease (Early symptomatic Alzheimer’s disease) who are apolipoprotein E ε4 (ApoE ε4) non-carriers with confirmed amyloid pathology. The regulators' review of Kisunla highlighted an iADRS slowing of 32 (donanemab vs placebo).

These drugs were studied in separate trials with different designs, populations, timepoints, and comparator arms. Leqembi's review focused on amyloid plaque reduction and the probability of superiority on ADCOMS, while Kisunla's review detailed iADRS and CDR-SB changes. The figures presented for each drug are from their respective trials and are not directly comparable. No implication of superiority of one drug over the other should be drawn from these separate findings.
