Skyrizi vs Stelara

IL-23-selective vs IL-12/23 inhibition, both reviewed for psoriasis and IBD

Not a head-to-head comparison. These drugs were studied in separate trials with different designs, populations and timepoints. Each figure below comes from that drug's own reviews and cannot be lined up against the other's.

What the regulatory reviews say

Skyrizi (risankizumab) is an IL-23 selective inhibitor reviewed for the treatment of active psoriatic arthritis (PsA) in adults, moderate-to-severe plaque psoriasis in adults who are candidates for systemic therapy or phototherapy, and moderate to severe plaque psoriasis in children and adolescents from the age of 6 years who are candidates for systemic therapy. One key finding from its review indicated a sPGA 0 or 1 response rate of 61%.

Stelara (ustekinumab) is an IL-12/23 inhibitor reviewed for adult patients with moderately to severely active ulcerative colitis, adult patients (18 years or older) with moderate to severe plaque psoriasis (Ps) who are candidates for phototherapy or systemic therapy, and moderate to severe Crohn’s disease. Its review noted a Clinical Response at Week 6 (Induction Study 3001) of 34.3% and a Clinical Response at Week 6 (Induction Study 3002) of 55.5%.

It is crucial to understand that these drugs were studied in separate trials with different designs, patient populations, endpoints, and timepoints. The figures presented are from each drug's own pivotal trials and are not head-to-head comparisons. Therefore, no direct comparison or implication of superiority between the two drugs should be made based on these figures.
