Caribou Biosciences, Inc. (CRBU) is an oncology research site. The company is developing vispa-cel, an allogeneic CAR-T cell therapy, which demonstrated an overall response rate (ORR) of 82% and a complete response (CR) rate of 67% in the ANTLER phase 1 trial (N=27). The median progression-free survival (PFS) for vispa-cel in this trial was 17.1 months. The ANTLER phase 1 trial included 16 patients in dose escalation, with dose levels of 40, 80, or 120 million CAR-T cells, and 63 patients in dose expansion for 2L LBCL. The recommended phase 2 dose (RP2D) for vispa-cel is 80 million CAR-T cells. Additionally, 6 patients with 3L+ LBCL who had prior CD19-targeted therapy were included in the ANTLER phase 1 trial. The ANTLER-3 trial has an enrollment of 250 patients.

Caribou Biosciences is also developing CB-011, another allogeneic CAR-T cell therapy. In the CaMMouflage phase 1 trial (N=12), CB-011 achieved an ORR of 92%, a complete response (CR) or stringent CR (≥CR) rate of 83%, and minimal residual disease (MRD) negativity in 91% of patients. The dose of CAR-T cells for CB-011 in this trial was 450 million cells. Dose levels for single dose in the CaMMouflage phase 1 trial included 150 million (N=6), 300 million (N=13), 450 million (N=13), and 800 million (N=3) CAR-T cells. The dose for dose expansion is 450 million CB-011 CAR-T cells.
